Chloroquine is a potent inhibitor of SARS coronavirus infection and spread

Background
Severe acute respiratory syndrome (SARS) is caused by a newly discovered coronavirus (SARS-CoV). No effective prophylactic or post-exposure therapy is currently available.

Results
We report, however, that chloroquine has strong antiviral effects on SARS-CoV infection of primate cells. These inhibitory effects are observed when the cells are treated with the drug either before or after exposure to the virus, suggesting both prophylactic and therapeutic advantage. In addition to the well-known functions of chloroquine such as elevations of endosomal pH, the drug appears to interfere with terminal glycosylation of the cellular receptor, angiotensin-converting enzyme 2. This may negatively influence the virus-receptor binding and abrogate the infection, with further ramifications by the elevation of vesicular pH, resulting in the inhibition of infection and spread of SARS CoV at clinically admissible concentrations.

Conclusion
Chloroquine is effective in preventing the spread of SARS CoV in cell culture. Favorable inhibition of virus spread was observed when the cells were either treated with chloroquine prior to or after SARS CoV infection. In addition, the indirect immunofluorescence assay described herein represents a simple and rapid method for screening SARS-CoV antiviral compounds.

The first SARS showed up in 2002. By 2005 they had figured out what they were dealing with and also found that they could treat it. Therefore, no vaccine was needed. Fauci spoke glowingly about the results when this research came out. Why did he turn his back on this research?

Why I’m kind of curious about stats from Africa or other tropical areas that use HCQS for other reasons.

Used the John Hopkins data to show the impact in India for one non-believer. they had the same approximate positive tests per million as the US and one tenth the death rate. Simple to see that the malaria drug treatments common in the population and largely HCQ were providing protection from cases becoming severe and causing death.

We’ve seen this all along and 45 just followed our lead in trying to get it followed.

Diet tonic water with quinine (Schwepps) plus favorite juice or SF drink mix. This has become my go to / daily drink and I am feeling a lot better. First I noticed better sleep, then the general feeling of malaise went away. Now I can focus better and I am starting a minor exercise routine.

My wife was exposed and contracted Covid and now has the anti-bodies. I never got it.

How many died because of this? Seriously.

There is widespread scorn for Cuomo over his nursing home policies and mishandling of it with a death count.

It pales in comparison to how many lives might have been saved with this cheap and dependable drug and our “expert bureaucrats” led a media charge to shit all over it.

Unforgivable.

Putting at risk people on HCQ as a preventative measure and treating with HCQ as soon as symptoms present would have stopped the virus and the pandemic in it's tracks

The Dean of Yale medical school has said that over 75% of the corona virus deaths could have been prevented if HCQ treatment had initiated at start of early symptoms or even later before major complications

Read this paper back when it was first published. It is a misnomer that HCQ only acts to potentiate the transport of zinc across the cell walls and increase pH in cells - it also interferes the cleavage of the ACE2 receptor which the CV-19 virus needs to be able to bind to the ACE2 receptors which is the primary mode of replication for the virus.

This was touted has a huge breakthrough for SARS at the time by Fauci and the rest of the medical community so it has been common knowledge in the medical community for 15 years.

HCQ was touted by Fauci as a safe and effective drug to prevent and treat SARS and other corona virus infections that absolutely should be taken because it was so safe and effective.

The whole demonization of HCQ as a preventative and treatment for CV-19 is inexplicable in any rational analysis and how and why the medical community played along is even more puzzling.

There is a large gap between showing that a compound works in cell culture, and demonstrating clinical efficacy of that compound.

There are many reasons this is so. The drug might be enzymatically altered after ingestion, so that it is not active for the intended purpose in the body. Or it might be eliminated from the body so fast that it never reaches the area where its needed in concentrations high enough to have an effect. Or it might display toxicities that make it unsafe to administer. Etc.

In cell culture, you have the luxury of placing the drug at desired concentrations directly on the cells without worrying about toxicity, metabolism, metabolic by-products, etc. But once you administer the drug to an animal, you might find it does not have the same effect at all that it had in cell culture.

In drug discovery efforts, hundreds of compounds are tested in order to try to determine which ones are worth further testing. You might find that out of 300 candidates that showed promise in cell culture, only 2 are worth testing further in animals and in clinical studies.

If I were looking at the efficacy of (hydroxy)chloroquine against coronaviruses (especially SARS-CoV-2), I would look at the recent literature, not papers published 15 years ago. What are the results of the recent clinical trials? Do they show efficacy? If not, then it's time to forget about (hydroxy)chloroquine as a potential treatment and focus on other drugs that have shown more promise.

Unfortunately, according to this recently published review, Chloroquine and hydroxychloroquine in the treatment of COVID-19: the never-ending story, rigorous studies have not shown that (hydroxy)chloroquine works.
