For a near-term historical example of unintended consequences of the long term use of a new drug, I suggest you review the history of Thalidomide. A drug intended for morning sickness, with extensive testing prior to its release, turned out to have devastating effects. It wasn't a malicious intent on the part of the drug companies, it was just the bad confluence of events that was never discovered in the initial study of the drug.
