Would DeSanctus have to sign the bill into law, or could he for purposes of discretion merely refrain from issuing a veto?

Perhaps.

Reading the law itself from this handout I linked to above this post, it appears clear as day that DeSantis would be exempt, yet this other reading I found of the law is as clear as mud and does not appear to list those same exemptions.
